Yom Kippur (YOHM ki-PAWR): Lit. Day of Atonement. A day set aside for fasting, depriving oneself of pleasures, and repenting from the sins of the previous year. Source: Glossary of Jewish Terminology
"And HaShem spoke unto Moses, saying: Howbeit on the tenth day of this seventh month is the day of atonement; there shall be a holy convocation unto you, and ye shall afflict your souls; and ye shall bring an offering made by fire unto HaShem. it's "Yohm kippoor" (day of atonement)
or "Yohm ha kippoorim" (day of the atonements)

Lots of wacky pronunciations out there.
YAWM ki-POOR is classical Hebrew, with the accent on the last syllable.
In the U.S. a lot of people -- especially those who haven't gone through rigorous religious training and learned pronunciation from a rabbi -- say YAHM (rhymes with "Mom") KIP-per (like a fish).
In some dialects of Yiddish it's been garbled even worse, into YING ki-POOR.
I'm not sure how it's supposed to sound in modern Hebrew. But judging by the fact that they've shifted the accent in most words from the last syllable to the penultimate (e.g. MITZ-vah instead of mitz-VAH), it's probably YAWM KI-poor in Israel.
The shorthand definition of Yom Kippur is "the Sabbath of all Sabbaths." As you say, you're not even allowed to do a lot of the stuff that is permitted on a normal Sabbath.

like yohm...but the "o" is short.
and the ashkenazis (especially in the u.s. with their american ashkenazi accent) say "MAZal tawf" or "MAZal tawv" instead of "maZAL tov"
